There are lots of coloring tutorials on the web that will walk you through the process step by step, but it's relatively simple to describe.

First you'll create a new blank layer under your black and white drawing layer. Set the drawing layer to blending mode multiply.

Second, use brushes, the pen tool, and whatever else you like to draw in color on the blank layer underneath. Play around with brush softness, texture, and line quality to get the effects that you like.

And yes, I agree with the idea of getting a graphics tablet, at some point. It is very fast to learn and makes a LOT of work easier. Still, it can still be done with a mouse, just a lot more carefully.
